    Abstract [en]

    The H295R in vitro cell line produces the majority of the steroidogenesis, for which reason it is commonly used as a screening tool for endocrine disrupting chemicals. Simultaneous determination of the precursor cholesterol and key steroid hormones could give a broad insight into the mechanistic disruption of the steroidogenesis. Steroid hormones have primarily been extracted from H295R incubation medium by means of liquid-liquid extraction (LLE) and the obtained recoveries and matrix effects have typically not been stated or assessed. In the present study a solid-phase extraction (SPE) method was developed and validated for the simultaneous extraction of cholesterol and five key steroid hormones pregnenolone, 17-hydroxyprogesterone, testosterone, cortisol and aldosterone from H295R incubation medium, and finally detected by LC-MS/MS. Cholesterol was recovered at a level of 55.7%, while steroid hormone recoveries ranged from 98.2 to 109.4%. Matrix effects varied between -0.6% and 62.8%. Intra-day precision was deemed acceptable, but the inter-day precision for pregnenolone and aldosterone exceeded the precision limit of 15% RSD. Although LLE has been the most frequently used extraction method in H295R studies, however, our investigation has shown that SPE may relatively easily extract and recover steroid hormones, potentially replacing LLE.

    Analytical methodologies based on liquid or gas chromatography coupled to tandem mass spectrometry for the simultaneous determination of two or more endogenous steroid hormones in human and animal plasma and serum has received increased attention the last few years. Especially in the clinical setting steroid profiling is of major importance in disease diagnostics. This paper discusses recent findings in such multi-steroid hormone procedures published from 2001 to 2012. The aim was to elucidate possible relationships between chosen analytical technique and the obtained analyte sensitivity for endogenous steroid hormones. By evaluating the success, at which the currently applied techniques have been utilized, more general knowledge on the field is provided. Furthermore the evaluation provides directions in which future studies may be interesting to conduct.

    This study demonstrates the addition of (14)C-cholesterol to the human cell line H295R will in-situ form radiolabeled steroid hormones allowing for new mechanistic and metabolic insights. The aim of the present study was to in-situ radiolabel steroid hormones from cell line-incorporated (14)C-cholesterol using the OECD guideline 456, H295R steroidogenesis in-vitro assay. Radiodetection of the steroid metabolites of the steroidogenic pathway allows for an improved understanding of the various enzymatic mechanisms involved without necessarily being dependent on quantification. Generated radiolabeled steroids were analyzed using HPLC hyphenated with a Flow Scintillation Analyzer (FSA). H295R cells were incubated with radiolabeled cholesterol and cell media were collected and prepared by solid phase extraction and analyzed with HPLC-FSA. For successful radiolabeling of the steroids in the steroidogenesis of H295R cells, radioactive cholesterol may potentially only need to be added just before the cells are incubated for 72h in well plates. Based on the obtained HPLC-FSA chromatograms, and confirmation of the observations by studies in the literature, a qualitative time profile for the production of steroid hormones was estimated. Multiple radiolabeled steroid hormones were identified by means of analytical standards and UV (ultraviolet) co-chromatography, though the elucidation of multiple metabolites remains unresolved. Although online radiodetection proved to suffer from suboptimal sensitivity, the concept of radiolabeling the steroidogenesis in H295R cells with (14)C-cholesterol and detecting the radiolabeled steroid hormones online was proved and may assist in further toxicological studies.

    One of the central characteristics of disciplines is that they create their own particular ways of knowing the world. This process is facilitated by the specialization and refinement of disciplinary-specific semiotic resources over time. Nowhere is this truer than in the sciences, where it is the norm that disciplinary-specific representations have been introduced and then refined by a number of different actors. As a consequence, many of the semiotic resources used in the sciences today still retain some traces of their historical roots.

    In this paper we analyse one such disciplinary-specific semiotic resource from the field of Astronomy—the Hertzsprung-Russell diagram. We audit the potential of this semiotic resource to provide access to disciplinary knowledge—what Fredlund et al (2012) have termed its disciplinary affordances. Our analysis includes consideration of the use of scales, labels, symbols, sizes and colour. We show how, for historical reasons, the use of these aspects in the resource may differ from what might be expected by a newcomer to the discipline.

    We suggest that some of the issues we highlight in our analysis may, in fact, be contributors to alternative conceptions and therefore propose that lecturers pay particular attention to the disambiguation of these features for their students.

    Becoming part of a discipline involves learning to interpret and use a range of disciplinary-specific semiotic resources (Airey, 2009). These resources have been developed and assigned particular specialist meanings over time. Nowhere is this truer than in the sciences, where it is the norm that disciplinary-specific representations have been introduced and then refined by a number of different actors in order to reconcile them with subsequent empirical and theoretical advances. As a consequence, many of the semiotic resources used in the sciences today still retain some (potentially confusing) traces of their historical roots. However, it has been repeatedly shown that university lecturers underestimate the challenges such disciplinary specific semiotic resources may present to undergraduates (Northedge, 2002; Tobias, 1986).

    In this paper we analyse one such disciplinary-specific semiotic resource from the field of Astronomy—the Hertzsprung-Russell diagram. First, we audit the potential of this semiotic resource to provide access to disciplinary knowledge—what Fredlund et al (2012) have termed its disciplinary affordances. Our analysis includes consideration of the use of scales, labels, symbols, sizes and colour. We show how, for historical reasons, the use of these aspects in the resource may differ from what might be expected by a newcomer to the discipline. Using the results of our analysis we then created an online questionnaire to probe what is discerned (Eriksson, Linder, Airey, & Redfors, in press) with respect to each of these aspects by astronomers and physicists ranging from first year undergraduates to university professors.

    Our findings suggest that some of the issues we highlight in our analysis may, in fact, be contributors to the alternative conceptions of undergraduate students and we therefore propose that lecturers pay particular attention to the disambiguation of these features for their students.

    In the late 70’s Gibson (1979) introduced the concept of affordance. Initially framed around the needs of an organism in its environment, over the years the term has been appropriated and debated at length by a number of researchers in various fields. Most famous, perhaps is the disagreement between Gibson and Norman (1988) about whether affordances are inherent properties of objects or are only present when they are perceived by an organism. More recently, affordance has been drawn on in the educational arena, particularly with respect to multimodality (see Linder (2013) for a recent example). Here, Kress et al. (2001) have claimed that different modes have different specialized affordances. Then, building on this idea, Airey and Linder (2009) suggested that there is a critical constellation of modes that students need to achieve fluency in before they can experience a concept in an appropriate disciplinary manner. Later, Airey (2009) nuanced this claim, shifting the focus from the modes themselves to a critical constellation of semiotic resources, thus acknowledging that different semiotic resources within a mode often have different affordances (e.g. two or more diagrams may form the critical constellation).

    In this theoretical paper the concept of disciplinary affordance (Fredlund et al., 2012) is suggested as a useful analytical tool for use in education. The concept makes a radical break with the views of both Gibson and Norman in that rather than focusing on the discernment of one individual, it refers to the disciplinary community as a whole. Put simply, the disciplinary affordances of a given semiotic resource are determined by those functions that the resource is expected to fulfil by the disciplinary community. Disciplinary affordances have thus been negotiated and developed within the discipline over time. As such, the question of whether these affordances are inherent or discerned becomes moot. Rather, from an educational perspective the issue is whether the meaning that a semiotic resource affords to an individual matches the disciplinary affordance assigned by the community. The power of the term for educational work is that learning can now be framed as coming to discern the disciplinary affordances of semiotic resources.

    In this paper we will briefly discuss the history of the term affordance, define the term disciplinary affordance and illustrate its usefulness in a number of educational settings.

    BackgroundThe thermal benefits of melanism in ectothermic animals are widely recognized, but relatively little is known about population differentiation in the degree of melanism along thermal gradients, and the relative contributions of genetic vs. environmental components into the level of melanism expressed. We investigated variation in the degree of melanism in the common frog (Rana temporaria; an active heliotherm thermoregulator) by comparing the degree of melanism (i) among twelve populations spanning over 1500 km long latitudinal gradient across the Scandinavian Peninsula and (ii) between two populations from latitudinal extremes subjected to larval temperature treatments in a common garden experiment.

    ResultsWe found that the degree of melanism increased steeply in the wild as a function of latitude. Comparison of the degree of population differentiation in melanism (PST) and neutral marker loci (FST) revealed that the PST > FST, indicating that the differences cannot be explained by random genetic drift alone. However, the latitudinal trend observed in the wild was not present in the common garden data, suggesting that the cline in nature is not attributable to direct genetic differences.

    ConclusionsAs straightforward local adaptation can be ruled out, the observed trend is likely to result from environment-driven phenotypic plasticity or ontogenetic plasticity coupled with population differences in age structure. In general, our results provide an example how phenotypic plasticity or even plain ontogeny can drive latitudinal clines and result in patterns perfectly matching the genetic differences expected under adaptive hypotheses.

    In 2009 we described an outbreak caused by extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli in southern Sweden that occurred during 2005–2006. An important finding from the investigation was the long carriage times of the ESBL-producing E. coli in several of the patients, which in some cases exceeded 30 months. Here we report findings from the continued follow-up of bacterial carriage. In September 2010, 5 of the 42 patients still carried the bacteria after a median of 58 months (range 41–59 months), 18 had had repeatedly negative cultures after shedding bacteria for a median of 7.5 months (range 0–39 months), 16 had died while still shedding the bacteria for a median of 9 months (range 0–38 months), and 3 had been lost to follow-up.

    BACKGROUND: Asthma genetics has been extensively studied and many genes have been associated with the development or severity of this disease. In contrast, the genetic basis of allergic rhinitis (AR) has not been evaluated as extensively. It is well known that asthma is closely related with AR since a large proportion of individuals with asthma also present symptoms of AR, and patients with AR have a 5-6 fold increased risk of developing asthma. Thus, the relevance of asthma candidate genes as predisposing factors for AR is worth investigating. The present study was designed to investigate if SNPs in highly replicated asthma genes are associated with the occurrence of AR.

    METHODS: A total of 192 SNPs from 21 asthma candidate genes reported to be associated with asthma in 6 or more unrelated studies were genotyped in a Swedish population with 246 AR patients and 431 controls. Genotypes for 429 SNPs from the same set of genes were also extracted from a Singapore Chinese genome-wide dataset which consisted of 456 AR cases and 486 controls. All SNPs were subsequently analyzed for association with AR and their influence on allergic sensitization to common allergens.

    RESULTS: A limited number of potential associations were observed and the overall pattern of P-values corresponds well to the expectations in the absence of an effect. However, in the tests of allele effects in the Chinese population the number of significant P-values exceeds the expectations. The strongest signals were found for SNPs in NPSR1 and CTLA4. In these genes, a total of nine SNPs showed P-values <0.001 with corresponding Q-values <0.05. In the NPSR1 gene some P-values were lower than the Bonferroni correction level. Reanalysis after elimination of all patients with asthmatic symptoms excluded asthma as a confounding factor in our results. Weaker indications were found for IL13 and GSTP1 with respect to sensitization to birch pollen in the Swedish population.

    CONCLUSIONS: Genetic variation in the majority of the highly replicated asthma genes were not associated to AR in our populations which suggest that asthma and AR could have less in common than previously anticipated. However, NPSR1 and CTLA4 can be genetic links between AR and asthma and associations of polymorphisms in NPSR1 with AR have not been reported previously.

    In many Western societies, there is a concern about the tendency of young people not choosing Science, Technology, Engineering, and Mathematics (STEM) education and occupations. In response, different initiatives have been launched. If one believes that science should have a place in more young people's lives, an important question is to what extent recruitment campaigns communicate messages that open up for STEM education to become relevant in young people's identity formation. Here, we analyse a Swedish government-initiated, primarily Internet-based recruitment attempt (‘The Broad Line Campaign’) aimed at increasing the number of young people choosing the natural science programme in upper secondary school. The campaign is based on marketing principles and deliberately draws on identity issues. The data analysed consists of campaign films and written resources describing the campaign. Data are analysed by use of the constant comparative approach in order to produce categories describing different messages about why to engage in STEM education. These messages are then analysed from an identity perspective using the concept of subjective values. Our results show that the messages communicated in the Broad Line campaign emphasise utility value, attainment value and relative cost rather than interest-enjoyment. The campaign communicates that the natural science programme is to be associated with a high attainment value without establishing relations to the field of science. Finally, potential consequences of the communicated messages in the campaign are discussed in light of previous research.

In our analysis we identify the following interpretative repertoires: 1) The securing competent labor repertoire, 2) The developing specific job skills or competences repertoire, 3) The bright future repertoire, and 4) The general increase interest in science repertoire. The results of this study may contribute to the self-reflection of industrial actors on how the choice of resources and repertoires may afford and constrain possibilities for breaking the cultural patterns of selection to STEM education.

    Young people's interest in pursuing science and science-intense educations has been expressed as a concern in relation to societal, economic and democratic development by various stakeholders (governments, industry and university). From the perspective of the scientific communities, the issues at stake do not necessarily correspond to the overall societal aims. Rather, initiatives to recruit young people to science are also ways for the scientific community to engage in the social and cultural reproduction of itself. For a community to survive and produce a future, it needs to secure regeneration of itself in succeeding generations. The aim of this study is to, from a perspective of social and cultural production/reproduction, shed light on an initiative from the scientific community to recruit young people to science education. This is a case study of one recruitment campaign called the Chemistry Advent calendar. The calendar consists of 25 webcasted films, produced and published by the science/technology faculty at a university. The analysed data consist of the films and additional published material relating to the campaign such as working reports and articles published about the campaign. The analysis focussed on what messages are communicated to potential newcomers. The messages were categorised by means of a framework of subjective values. The results are discussed both from a perspective of how the messages mirror traditions and habits of the scientific community, and in relation to research on students' educational choices.

    Abstract [en]

    Translating policies about sustainable development as a social process and sustainability outcomes into the real world of social-ecological systems involves several challenges. Hence, research policies advocate improved innovative problem-solving capacity. One approach is transdisciplinary research that integrates research disciplines, as well as researchers and practitioners. Drawing upon 14 experiences of problem-solving, we used group modeling to map perceived barriers and bridges for researchers' and practitioners' joint knowledge production and learning towards transdisciplinary research. The analysis indicated that the transdisciplinary research process is influenced by (1) the amount of traditional disciplinary formal and informal control, (2) adaptation of project applications to fill the transdisciplinary research agenda, (3) stakeholder participation, and (4) functional team building/development based on self-reflection and experienced leadership. Focusing on implementation of green infrastructure policy as a common denominator for the delivery of ecosystem services and human well-being, we discuss how to diagnose social-ecological systems, and use knowledge production and collaborative learning as treatments.

    Abstract [en]

    Teacher-researcher collaborations inevitably dissolve the border between who is teaching and who is researching, which in turn pushes discussions on research ethics as well as how to define, measure and ensure research validity. In an attempt to deepen the discussion on validity and ethical issues in teacher-research collaboration in science education research, this proposal seeks to disentangle how the notion of ‘ownership’ is transformed in various forms of teacher-researcher collaborations. The paper builds on data from three recent research projects in science education, representing different forms of teacher-researcher collaborations. The analysis focuses how the teachers’ and researchers’ ownership is played out in four stages of the research process; problem stating, design and implementation, analysis, and dissemination. The results indicate that ownership becomes actualised and altered in different ways, and at different stages, depending on the form of collaboration. A conclusion is that ownership needs to be accounted for in in all stages in the research process in order for science education research to accomplish deep relevance and justified implications. Since the question of ownership appears to be particularly troublesome in the last stages of the research process, a suggestion for future teacher-researcher collaboration is to plan for the project’s end, for example in terms of the participants’ anonymity and time to take part in analysis and dissemination of the results, already at the beginning.

    The latitudinal diversity gradient predicts higher species richness at lower latitudes. Here, we utilize the data from a long-term monitoring with malaise traps to analyse if spider communities in Sweden are affected by geographic gradients and if these effects hold independent of forest type. The species richness and the effective number of species in spider communities were not significantly related to the latitudinal gradient. The effective number of species and the taxonomic distinctness of spider communities were related to longitude, with a higher number, but fewer related species in western parts of Sweden. The species and family composition were significantly related to latitude independent of forest type, with a dominance of Linyphiidae individuals and species in the north. Our study demonstrates the suitability of malaise trap sampling to contribute to a better understanding of local spider communities, as several rare and locally new species were recorded in this study.

    The capacity of migratory species to adapt to climate change may depend on their migratory and reproductive strategies. For example, reproductive output is likely to be influenced by how well migration and nesting are timed to temporal patterns of food abundance, or by temperature variations during the brood rearing phase. Based on two decades (1988–2009) of waterfowl counts from a boreal catchment in southern Finland we assessed how variation in ice break-up date affected nesting phenology and breeding success in two sympatric duck species, Mallard Anas platyrhynchos and Eurasian Teal Anas crecca. In Fennoscandia these species have similar breeding habitat requirements but differ in migration distance; Teal migrate roughly seven times as far as do Mallard. Annual ice break-up date was used as a proxy of spring ‘earliness’ to test the potential effect of climate change on hatching timing and breeding performance. Both species were capable of adapting their nesting phenology, and bred earlier in years when spring was early. However, the interval from ice break-up to hatching tended to be longer in early springs in both species, so that broods hatched relatively later than in late springs. Ice break-up date did not appear to influence annual number of broods per pair or annual mean brood size in either species. Our study therefore does not suggest that breeding performance in Teal and Mallard is negatively affected by advancement of ice break-up at the population level. However, both species showed a within-season decline in brood size with increasing interval between ice break-up and hatching. Our study therefore highlights a disparity between individuals in their capacity to adjust to ice break-up date, late breeders having a lower breeding success than early breeders. We speculate that breeding success of both species may therefore decline should a consistent trend towards earlier springs occur.

    We studied time budgets and foraging methods in pre-breeding Mallard Anas platyrhynchos, (Eurasian) Teal Anas crecca, Wigeon Anas penelope, Pintail Anas acuta, Shoveler Anas clypeata and Gadwall Anas strepera in subarctic Norway in May. Among all six species studied, foraging accounted for the most common use of time, ranging from 19 % in male Pintail to 40–60 % in female Mallard, Teal, Pintail and Gadwall. Comfort behaviours amounted to 20–34 % of the time budget, and interaction and disturbance were marginal. Vigilance time ranged from 8 % in female Mallard to 20 % in male Pintail. Movement amounted to some 20 % of the time in most species and sexes. In Wigeon, sexes did not differ in time use, whereas in Mallard, Pintail and, in particular, Teal, females foraged more and engaged less in vigilance and interactions than did males. In addition, Teal and Mallard males engaged in the riskier foraging methods less than females, but more in those permitting vigilance. Although overlap in feeding methods was large among these species, Mallard and Teal were generalists, feeding at all depths, Wigeon foraged mainly in shallow water and Pintail foraged essentially in deep water. Our results support the income/capital breeder hypothesis with respect to males only; compared to lighter species, heavier species allocated less time to foraging but more to vigilance. We found no support for the hypothesis that long-distance migrants forage more to compensate for energy loss due to migratory flight. Foraging time in females was related to breeding phenology; early nesters spent more time feeding than later nesters.

Surprisingly, this period has seldom been studied. To start filling this gap for dabbling ducks and especially Eurasian Teal Anas crecca, we studied their foraging time and foraging methods along their Western European flyway from wintering to breeding grounds. Differences in foraging activity between sexes, species, years, seasons, sites and diel patterns were checked, as well as potential effects of disturbances by potential predators. Ducks of both sexes presented a fairly constant nocturnal foraging along the flyway, whereas diurnal foraging increased at periods of high requirements (spring, breeding, moulting). Ducks might thus not only be income breeders, but also income migrators (relying on the food they encounter along their flyway to fuel their travel). Moreover, Teal foraging depth increased along the flyway when disturbance due to potential predators decreased. This probably reflects their diet switch (from granivorous to carnivorous). The decrease in predator pressure probably makes it possible for teals to use riskiest behaviors (greater depths associated to eyes underwater and less prevention of predators).We thus highlight the need for adequate management of staging wetlands all along the flyway, since ducks have to fulfill high energy requirements there. Water level control may be useful to provide more shallow foraging habitats in which they can use less risky foraging techniques at times when nutrient needs increases and predation risk is relatively high.

    Very little is still known about the ecology of dabbling ducks on staging areas in spring, despite the fact that this is a crucial period of the year when birds refuel during migration and prepare for subsequent reproduction. This lack of knowledge translates into the current inability to predict the consequences of changes in the environment and harvesting for the population dynamics of ducks. We have launched a joint project to study dabbling ducks in the flyway connecting wintering grounds in France with breeding areas in Sweden. The aim is to study individual dabbling ducks in a circumannual perspective. We are assessing food resource limitation, density-dependent effects and the role of competition by estimating available food, by using a reference functional response curve and by recording the foraging behaviour of wild individual dabbling ducks. We also study survival probability and breeding success, and we will assess inter-individual variation. Birds are captured, weighed, marked and subsequently observed in order to study foraging behaviour and potentially measure the changes in body mass across time through recaptures. Ducks are fitted with nasal marks for individual recognition. Eventually, we will provide a model to predict the number of young that a female duck will produce after simple measurements such as its date of arrival on the breeding grounds and its body-mass at this date. Duckling foraging efficiency and survival will be measured and the relationship with corresponding estimates for their mother will be explored.

    Ocean acidification (OA) can shift the ecological balance between interacting organisms. In this study, we have used a model system to illustrate the interaction between a calcifying host organism, the blue mussel Mytilus edulis and a common bivalve bacterial pathogen, Vibrio tubiashii, with organisms being exposed to a level of acidification projected to occur by the end of the 21st century. OA exposures of the mussels were carried out in relative long-term (4 months) and short-term (4 days) experiments. We found no effect of OA on the culturability of V. tubiashii, in broth or in seawater. OA inhibited mussel shell growth and impaired crystalline shell structures but did not appear to affect mussel immune parameters (i.e haemocyte counts and phagocytotic capacity). Despite no evident impact on host immunity or growth and virulence of the pathogen, V. tubiashii was clearly more successful in infecting mussels exposed to long-term OA compared to those maintained under ambient conditions. Moreover, OA exposed V. tubiashii increased their viability when exposed to haemocytes of OA-treated mussel. Our findings suggest that even though host organisms may have the capacity to cope with periods of OA, these conditions may alter the outcome of host–pathogen interactions, favouring the success of the latter.

    Abstract [en]

    Background - When sepsis or bacteraemia is suspected, patient blood samples are cultivated in blood culture bottles and then further incubated for identification of the organism and antimicrobial susceptibility testing. These methods are slow, identifying causative pathogens in a couple of hours, and antibiotic susceptibility results within 18-36 hours.

    Here we present optimization of several parameters in order to evaluate if the MBT ASTRA™ method can be a rapid tool, used for routine antibiotics susceptibility testing, in a clinical laboratory.

    Methods – MALDI-TOF MS measurements were performed with a Microflex LT/SH bench-top mass spectrometer (Bruker) with standard settings. The resulting spectra were uploaded in the MBT-ASTRA™ software, which normalizes the peaks and determines the AUC and RG values for each setup.

    Results - The bacterial preparation steps generated a new protocol, which reduced time with 30-60 minutes.

    The antibiotics susceptibility test was optimized for 90 minutes incubation time. 200 µl McFarland 0.5 bacterial suspension in broth were incubated in broth at 37°C, with and without 32 µg/ml Cefotaxime, 16 µg/ml Meropenem and 4 µg/ml Ciprofloxacin.

    The suspensions were transferred to 0.45 µm pore size filter membraned 96 well plate. They were centrifuged; washed; fixated and eluted; put on a MALDI-target, and covered by matrix solution. All could be automated with robot, which reduced time with 60 minutes.

    Conclusion – Rapid susceptibility testing becomes more requested with the increase of resistance bacteria causing infections. Our study can be a valuable tool for clinical laboratories striving for reduction in time handling of antibiotic susceptibility testing.

    Antibiotics in the environment are a potential threat to environmental ecosystems as well as human health and safety. Antibiotics are designed to have a biological effect at low doses, and the low levels detected in the environment have turned focus on the need for more research on environmental occurrence and fate, to assess the risk and requirement for future regulation. This article describes the first occurrence study of the antibiotic polyether ionophores (lasalocid, monensin, narasin, and salinomycin) in the Danish environment. Various environmental matrices (river water, sediment, and soil) have been evaluated during two different sampling campaigns carried out in July 2011 and October 2012 in an agricultural area of Zealand, Denmark. Lasalocid was not detected in any of the samples. Monensin was measured at a concentration up to 20 ng·L−1 in river water and 13 µg·kg−1 dry weight in the sediment as well as being the most frequently detected ionophore in the soil samples with concentrations up to 8 µg·kg−1 dry weight. Narasin was measured in sediment samples at 2 µg·kg−1 dry weight and in soil between 1 and 18 µg·kg−1 dry weight. Salinomycin was detected in a single soil sample at a concentration of 30 µg·kg−1 dry weight.

    A multi-residue pressurised liquid extraction (PLE) methodology has been established for the determination of the four ionophores: lasalocid, monensin, salinomycin and narasin in solid environmental matrices. The PLE methodology is combined with solid phase extraction as clean-up using liquid chromatography coupled to tandem mass spectrometry applying electrospray ionisation for detection. The samples were freeze-dried prior to extraction. The absolute recoveries for soil and sediment ranged from 71 to 123% (relative standard deviation (RSDs) below 16%) and in the range 94–133% (RSDs 9–35%) for poultry manure. The final method allowed for the detection of four ionophores down to a few hundred ng kg−1 in natural solid matrices with limit of quantifications (LOQs) being 0.96, 0.87, 0.98, and 0.64 μg kg−1 in soil for lasalocid, monensin, salinomycin, and narasin, respectively. Corresponding LOQs in sediment were 1.28, 1.34, 1.39, and 0.78 μg kg−1 for the respective ionophores, while in manure the LOQs were 0.98, 1.01, 1.45, and 1.01 μg kg−1.

    Syftet med denna studie är att få en bättre förståelse för hur fysisk aktivitet kan användas i skolan för att skapa mer gynnsamma förutsättningar för elevernas lärande i matematik. I studien har det undersökts om det finns en effekt av fysisk aktivitet på lång sikt på elevernas koncentration, arbetsminne och kunskaper i matematik av ett återkommande kort pass med fysisk aktivitet. Studien genomfördes med 175 elever i årskurs 7 på fyra olika skolor. Ett inledande pass på sju minuter med pulshöjande övningar introducerades i fem av tio grupper i början på matematiklektionen. Resultaten visar en förbättring av elevernas koncentration och arbetsminne i interventionsgruppen i förhållande till kontrollgruppen. Elever med olika kön och prestationsnivå påverkas på samma sätt av den fysiska aktiviteten. Det finns inte heller någon skillnad i hur elever som kommer från olika skolkontext påverkas av den fysiska aktiviteten.

    We describe the design background of the mathematics game Fingu for iPad aimed at 4 to 8 year old children. We first describe how Fingu theoretically can support children's development of fundamental arithmetic skills, focusing on conceptual subitizing, the embodiment of numerosity, and finger gnosis. Then we present the results of an exploratory micro-longitudinal study of the game with 11 5- and 6-year old children playing the game for several weeks and being filmed at three occasions. We discuss how their behavior with the game develops over time and can be related to the development of arithmetic skills. Finally we discuss how we will proceed testing the effectiveness of Fingu in a larger controlled study.

    Apicidin is a potent histone deacetylase inhibitor (HDACI) that selectively binds to histone deacetylases (HDACs) class I and interferes with the deacetylation process, which results in modification of acetylation level of cellular proteins. The aim of the study was to investigate the potential time and dose dependent cytotoxicity of the test compound, Apicidin, in pancreatic cancer cells Capan-1 and Panc-1 as well as estimate maximal tolerable dose (MTD) of the test agent and determine EC50 using four complementary colorimetric cytotoxicity or viability assays. The cells were treated with increasing concentrations of Apicidin (0-5000nM) for 2, 4 and 6h (short term exposure) or 24, 48 and 72h (long term exposure) before conducting cytotoxic analyses with lactate dehydrogenase assay or viability analyses with sulforhodamine B (SRB), methyl tetrazolium (MTT) and crystal violet (CV) assays. In order to investigate whether Apicidin irreversibly affects the cells already during the short term exposure, the medium containing Apicidin was removed and replaced with fresh culturing medium after 6h of treatment. The cells were then incubated for additional 24, 48 or 72h before carrying out the analysis. The results obtained from cytotoxicity and viability assays indicated, that Apicidin was well tolerated by both cell lines at concentrations below 100nM at any given time point and at all applied concentrations during the short term (6h or less) treatment. Continuous prolonged term exposures (48h or greater) of the cells to Apicidin with concentration exceeding 100nM resulted in significantly increasing cytotoxicity and sustained significant loss of cell viability. Moreover, long term exposure of pancreatic cancer cells Capan-1 and Panc-1 to Apicidin concentrations exceeding 100nM showed an initial anti-proliferative effect before cytotoxicity onset. In summary, MTD was exposure time dependent and estimated to 100nM for long term treatment and to at least 5000nM for treatment not greater than 6h. EC50 concentration of Apicidin was established after long term treatment, however with some variation when comparing the different assays and cell lines. Results from this study may encourage reinvestigating the capacity of potent HDACI Apicidin as an attractive agent for interfering with the deacetylation process catalyzed by HDACs for potential pancreatic cancer intervention.

    Childhood collecting in nature: quality experience in important places2016Konferansepaper (Fagfellevurdert)

    There is growing concern for both the decline of direct experience people have with nature, and the quality of that experience. This diminished experience has negative consequences for public awareness and concern for biodiversity and ecosystem health (Miller, 2005). At the same time, a diminished experience of nature appears to have a substantial negative impact on child development (Kahn & Friedman, 1995; Kahn, 2002; Matteo, Barthel, & Lars, 2014; Pyle, 1993; Thomashow, 2002). These concerns are heightened in the urban context where increased urbanization shows a relationship with a reduction in biodiversity and ecosystem health (MA, 2005; Sala et al., 2000). Additional concern comes from studies showing decreasing ecological knowledge among growing urban populations (McDaniel & Alley, 2005; McKinney, 2002). In an attempt to address these concerns and contribute toward a better understanding of the importance of childhood experience of nature, this study investigates one specific example, collecting in nature. Studies show that childhood collecting in nature (the gathering of rocks, shells, feathers, etc. as part of play and free exploration) is a widespread phenomenon (Lekies & Beery, 2013), and yet, very little is understood about this behavior. This study explored the details of childhood collecting in nature with an emphasis on the places of this experience. Participants consisted of a random sample of undergraduate students at a Swedish university (N = 380) participating in a survey focused upon early life outdoor experiences. Responses included multiple choice and Likert scale items, along with data from open-ended questions. In addition, participants were invited to discuss their experience of childhood collecting in greater detail via a semi-structured interview. Fourteen interviews were conducted as follow-up to the survey. Data review considered descriptive statistics, correlations, and regression analysis triangulated with the qualitative data from the open-ended responses and interviews. Results highlight the importance of specific places in the childhood experience of nature, the importance of nearby nature, and further, provide preliminary support for a model for environmental concern (Wolf-Watz, 2015). Ultimately, the study illuminates the idea of childhood development as a cultural ecosystem service and provides implications for nature-based solutions, such as green infrastructure, to support childhood nature experience.

    Outdoor educators are concerned about a perceived human disconnection from nature. There is awareness of a lack of human affiliation, connection, or identity with nonhuman nature and its impact on attitudes and behaviors. This essay raises the possibility that despite our concern, we may contribute toward this disconnection via language that supports a separation of the natural and the cultural. Our ability to separate ourselves conceptually from the rest of nature may be partially to blame for environmental degradation, therefore challenging the nature-culture dichotomy is both useful and constructive. This essay will present examples of how outdoor educators can attempt to get past this problematic dichotomy and motivate more relational discourse within the practice of outdoor education.

    Given concerns for a severely diminished childhood experience of nature, coupled with alarm for a rapidly diminishing global biodiversity, this article considers the potential for childhood nature experience to be an important part of biodiversity understanding. Findings from two studies are integrated and presented as windows into childhood nature experience to illuminate important aspects of sensory rich learning. In one study from Sweden, semi-structured interviews with adults were conducted and analyzed to explore an understanding of the sensory experience of childhood collecting in nature via participant memories. In the second study, direct observations of children's play and exploration in an outdoor kindergarten in Norway were conducted and analyzed. Bringing these two studies together for shared analysis is useful for investigating biodiversity experience and understanding. Analysis supports the idea that the experience of biodiversity, actual childhood interaction with variation and diversity with living and nonliving items from nature allows children important learning opportunities, inclusive of biodiversity understanding. The results support practical implications for sensory rich environmental education and underscores the practical importance of childhood access to nature.

    This study investigates outdoor recreation participation within a multifunctional landscape, a UNESCO Man and Biosphere Reserve. The reserve, the Kristianstad Vattenrike located in southern Sweden, has made a deliberate effort to make the experience of biodiversity possible for residents and visitors. Recreation is a keypart of the biodiversity conservation effort in the area, represented by the infrastructure of the Kristianstad Vattenrike's 21 visitor sites. Given the biosphere reserve context, this study investigates the question of whether there is a relationship between outdoor recreation participation and place attachment. Survey data was collected using concurrent application of multiple sampling strategies including both probability and purposive sampling of local adult residents of the biosphere area. Quantitative analysis showed a significant positive relationship between the level of outdoor recreation participation and place attachment. Qualitative data supported this relationship with more details about place attachment within the studied area. The study confirms a relationship between place attachment and outdoor recreation and provides insight into how the biosphere reserve context supports this relationship. The results of this study show that significant biodiversity management in close conjunction with outdoor recreational opportunity can be achieved and provides opportunities for human engagement and experience of biodiversity.

    Management Implications: This research can help managers design recreational settings that support biodiversity conservation goals. Our research found that:

    • A leading motivation for outdoor recreation participation is nature experience and this motivation can be used by managers to highlight a biodiversity conservation interpretive message in the design of outdoor recreation infrastructure.

    • Providing proximate access to nature based outdoor recreation, to support deliberate and direct experience of biodiversity, is an important component of engaging the public in biodiversity conservation.

    • Recreation proximity alone will not create public engagement in biodiversity conservation. However,proximity as a part of a deliberate institutional design including biodiversity conservation, sustainable development, and logistic support for research and monitoring may be critical for public engagement.

    En stor del av Biosfärområdet Kristianstad Vattenrikes verksamhet handlar om att ge människor möjlighet att uppleva den biologiska mångfalden i biosfärområdet, med syfte att skapa intresse och motivation att bevara och förvalta de unika naturvärdena. De samhällsvetenskapliga projekt som beskrivs i denna artikel har en stark anknytning till detta mål.

    Abstract [en]

    This study investigates the visitor experience at a Swedish nature center within aUNESCO biosphere reserve. The question of whether this interpretive facility succeedsin motivating the visitor to get outdoors for a direct experience of nature is explored. Useof the environmental connectedness perspective and concerns about diminished natureexperience support the importance of this study. A number of qualitative methodologiesare used to investigate the research questions, including thought listing, phenomenology,and field observation. Results indicate that this particular nature center generallysucceeded in the goal of inspiring visitors for a direct nature experience. The success inmotivating visitors appears to be a result of a number of key variables, including placebasedexhibitry, access, and personal visitor factors. Given the setting for this study, weconclude that interpretive nature centers have the potential to play an important role inthe re-imagination of urban environments.

    Abstract [en]

    Objective: The objective of this study is to explore the co-evolutionary foundation for place-based human affiliation with nonhuman nature, and its potential to support sustainable development at the local level. In particular, we analyse the Topophilia Hypothesis, an expansion of the Biophilia Hypothesis which includes also non-living elements in the environment.

    Methods: The study represents a multidisciplinary conceptual analysis of how biological selection and cultural learning may have interacted during human evolution to promote adaptive mechanisms for human affiliation with nonhuman nature via specific place attachment. 

    Results and Conclusions: The Biophilia Hypothesis has been one of the most important theories of human connectedness with nature, suggesting a genetically based inclination for human affiliation with the biological world. The Topophilia Hypothesis has extended the ideas of Biophilia to incorporate a broader conception of nonhuman nature and a co-evolutionary theory of genetic response and cultural learning. It also puts more emphasis on affiliation processes with the local environment. We propose that nurturing potential topophilic tendencies may be a useful method to promote sustainable development at the local level, and ultimately at the global level. Tendencies of local affiliation may also have implications for multifunctional landscape management, an important area within sustainability research, and we provide some examples of successful landscape management with a strong component of local engagement. Since human affiliation with nonhuman nature is considered an important dimension of environmental concern and support for pro-environmental attitudes, the Topophilia Hypothesis may provide a fruitful ground for a discourse within which scholars from many scientific fields, including human evolution and humanistic geography, can participate.

    Human affiliation with nonhuman nature is an important dimension of environmental concern and support for pro-environmental attitudes. A significant theory of human connectedness with nature, the Biophilia Hypothesis, suggests that there exists a genetically based inclination for human affiliation with the biological world. Both support and challenge to the Biophilia Hypothesis are abundant in the literature of environmental psychology. One response that both challenges and builds upon the Biophilia Hypothesis is the Topophilia Hypothesis. The Topophilia Hypothesis has extended the ideas of biophilia to incorporate a broader conception of nonhuman nature and a co-evolutionary theory of genetic response and cultural learning. While the Topophilia Hypothesis is a new idea, it is built upon long-standing scholarship from humanistic geography and theories in human evolution. The Topophilia Hypothesis expands previous theory and provides a multidisciplinary consideration of how biological selection and cultural learning may have interacted during human evolution to promote adaptive mechanisms for human affiliation with nonhuman nature via specific place attachment. Support for this possible co-evolutionary foundation for place-based human affiliation with nonhuman nature is explored from multiple vantage points. We raise the question of whether this affiliation may have implications for multifunctional landscape management. Ultimately, we propose that nurturing potential topophilic tendencies may be a useful method to promote sustainable efforts at the local level with implications for the global.
